AACR Special Conference in Cancer Research: Targeting RAS—Mechanisms, Novel Therapeutics, and Clinical Impact

For decades, RAS was considered one of cancer biology’s most important and challenging therapeutic targets. Today, the field stands at a pivotal moment. The emergence of direct RAS inhibitors, novel combination strategies, and a deeper understanding of RAS signaling networks has transformed what was once viewed as an “undruggable” pathway into a rapidly evolving therapeutic landscape.

Bringing together leaders from academia, industry, and clinical oncology, the conference will explore how advances in structural biology, chemical biology, omics, and immunology continue to shape our understanding of RAS-driven cancers and guide therapeutic approaches. Special emphasis will be placed on emerging therapeutic vulnerabilities, mechanisms of response and resistance, and opportunities to improve outcomes across multiple tumor types, including pancreatic, colorectal, lung, melanoma, and hematologic malignancies. 

As RAS-targeted agents enter clinical practice and new generations of inhibitors advance through development, fundamental questions remain regarding resistance, pathway adaptation, tumor heterogeneity, and optimal therapeutic combinations. A dedicated session will spotlight promising clinical trial discoveries from submitted abstracts that have the potential to impact future therapeutic strategies.
